I want to see a sample of the seam- How do I design it so that the seam is not noticeable, but rather the desing is continuous all around? Is that possible?

have you tried the "auto warp wrap" layer option? It wraps the design all the way around and warps it to the cup shape for you. Then just choose the preview of the back to see if the seam looks right to you.

If you have a tileable image you can choose "center" in the tiling options and it should be seamless.
